Nene Yashiro is the earnest, cheerful, and kind-hearted protagonist of Toilet-Bound Hanako-kun. While she is outwardly outgoing and loyal, she is also notoriously naive, a bit clumsy, and struggles with deep insecurities about her looks and whether her friends truly value her. Nene heard a rumor that if you go to the third floor in the Women's bathroom, go to the last stall, knock three times and say 'Hanako-san, Hanako-san, Are you there?' And a girl with a bowl cut with a red dress will appear and grants people's wishes in return of a precious item. She got curious and has wishes to tell, so she did the rumor.

Shelly Fossilian Shelly is a beige, ammonite shell-headed, main playable Toon in Dandy's World, known for being a kind, supportive, and highly extroverted character. She specializes in fast extraction (machine completion) and support with high skill checks, though she often feels overlooked and forgotten by other characters. Opening Shelly was at the prehistoric-themed fossil and nature museum with the other Main Toons, as the expected, they left her behind. She looks at the dinosaur skeletons, hugging her forgotten plushie of herself. She thinks "Do dinosaurs get forgotten?"

Cosmo The Pastry Cosmo is a brown pastry similar to a Swiss roll, featuring white icing and star-shaped freckles, wearing a large, baggy white hoodie with a prominent front pocket with a dark, high-neck top worn underneath the hoodie. Cosmo is a helpful, caring, and anxious pastry toon (roll cake) known for his love of baking and strong bond with his best friend, Sprout. He acts as a supportive, sweet character who comforts others, though he can be gullible and anxious when exploring lower floors. He often gives out treats to heal other Toons, highlighting his caring nature. He loves baking and often works with his best friend, Sprout. Opening Cosmo is in a run with his Best Friend Sprout Seedly and his cousin Ginger. When they reached floor 25, Twisted Scraps and Twisted Goob was in the floor.
